Research focus

  • Examination of the mechanisms of metal nanoparticles interaction with cells of pro- and eukaryotic organisms in vitro and in vivo.
  • Studies of morphology and replication of viruses, bacteria and other microorganisms.
  • Electron microscopic studies of structures in tears in patients with ophthalmic diseases.
  • Examination of reproduction and antitumor activity of oncolytic viruses in vitro and in vivo.
  • Visualization of macromolecular constructions and their interaction with cells.

Main scientific results

  •  Mechanisms of oncolytic activity of vaccinia virus recombinant producing apoptin were understood using carcinoma A431 xenografts in nude mice. Cytoplasmic effect of apoptin, enhancing vaccinia virus oncolytic effect was found.
  • The structures were visualized and examined in lachrimal fluid of patients suffering from glaucoma, diabetic retinopathy and macular dystrophy. Morphological features of vitreous body and internal limiting membrane were described in patients with macular dystrophy.
  • Plasmonic effect of gold nanorods under laser irradiation was examined and its damaging effect of melanoma B16 in mice was shown.
  • Cells of hepatoma A-1 and peritoneal macrophages possess identical mechanisms of internalization of gold nanoparticles in vitro and in vivo. The cells use different mechanisms of endocytosis, specific to the type of cells, for internalization of nanoparticles.
  • The method of preparation of palladium nanoparticles, identical to released from car motors in atmosphere, for application in studies with cell cultures has been elaborated. The penetration of palladium nanoparticles through the plasma membrane in MDCK culture cells and their accumulation in the nucleus was found. The macrophages internalized palladium nanoparticles by phagocytosis and clathrin-mediated endocytosis, and the nanoparticles accumulated in lysosomes. Thus, palladium nanoparticles inhaled from atmosphere, could damage epthelium of airways and accumulate in alveolar macropahges.
  • Subsequent stages of the damaging effect of artificial ribonucleases on vaccinia virus, influenza, and bee paralysis virus were visualized. Development of viral envelope alteration and damage of viral nucleoid was observed.
